One form, Alzheimer's disease, is likely due to a mix of age-related brain changes, genetics, overall health, and lifestyle. Key genetic variants include the ApoE4 allele and Clusterin gene risk alleles. These increase the risk of Alzheimer's, with ApoE4 carriers facing up to twelve times the risk compared to common variants.

Abnormal protein deposits, amyloid plaques and tangles in Alzheimer's, and Lewy bodies in Lewy body dementia, damage nerve cells. Different types of dementia correspond to specific brain damage in various areas. Other factors like traumatic brain injury, certain infections such as HIV, and high alcohol consumption may also contribute to dementia. Vascular dementia, caused by disrupted blood flow due to damaged blood vessels, is another prevalent form.
